A xenograft model of metastatic human melanoma was developed to compare melanoma lines with and without experimental metastases. This model compares the response of human melanoma to treatment in vivo in melanoma cells, which have either spontaneously developed metastases or were implanted into mice as human melanoma lymph node metastases. A single thymidine kinase-expressing transfected melanoma cell line, A375SM, was grown in either nude mice alone or in combination with human lymph node metastases derived from a patient with metastatic melanoma. Tumor growth was measured, and histologic features were documented. Spontaneous metastases, which were most common in the brain, lymph node, and lungs, were significantly increased in mice coimplanted with A375SM tumor cells and lymph node metastases compared with mice coimplanted with A375SM tumor cells alone (P
